該載體通過(guò)自殺基因表達與否篩選陽(yáng)性重組子。當載體與片段連接成功時(shí)，自殺基因無(wú)法正確表達，包含重組子的細胞可以正常生長(cháng)；當載體與片段連接不成功時(shí)，自殺基因正確表達，包含載體的細胞無(wú)法生長(cháng)，即“Zero”背景。適用于平端克隆。
